André Alexandre Grangeia Gouveia, completed both the Master degree in Microbiology and Bachelors degree in Biology at Aveiro University, Biology department. At the moment he is working on its PhD fellowship at ITQB NOVA - Instituto de Tecnologia Química e Biológica António Xavier. His mains scientific interests are microbial ecology of extreme habitats, focusing in bacterial extremophiles. Particularly, he is interested in understanding the cellular function that enables tolerance/resistance of extremophile bacteria and its application has a tool to solve major global challenges. Currently, he is working with a model organism for radiation resistance, Deinococcus radiodurans to unveil the mechanisms of resistance to radiation conditions as well as their ability to cope with heavy-metals. His aim is to use Deinococcus species (Deinococcus radiodurans and Deinococcus indicus) has a tool for heavy-metal detoxification of waste water systems. At the moment has aquired expertise in in the design of experimental protocols for D. radiodurans in (1) Structural characterization, including protein expression, purification, crystallization and structure determination; (2) Microscopy, including scanning electron microscopy, transmission electron microscopy as well as Cryo electron microscopy sample preparation (workshop) and negative staining; (3) Biological assays, including oxidative stress response and ionizing radiation survival assays.

2020/11/23 - 2020/11/26 A short course of online lectures will provide an overview of the approaches and methods used in electron cryo-tomography. Topics to be introduced include basic TEM/STEM optics and operation, methods of sample preparation, lamella preparation and serial surface imaging using the focused ion beam – scanning electron microscope (FIB-SEM), correlative fluorescence microscopy (CLEM), and 3D reconstruction of tilt series tomograms. The course should serve as a useful background for practical hands-on workshops.

2023/01/15 - 2023/01/27 Through the IMpaCT European Project, I had the opportunity to learn and perform Cryo-Scanning transmission electron microscopy (STEM) coupled with Energy-dispersive X-ray spectroscopy (EDX) to unveil the metal composition of Deinococcus radiodurans and Deinococcus Indicus polyphosphate granules. During this period of time I was trained to do sample preparation, acquire data of STEM and STEM-EDS. and data processing and analisis using VELOX software. 2022/01/19 - 2022/01/24 I had the opportunity to travel to the ESRF to learn and perform analysis of the elemental distribution of D. radiodurans cells using the the ID16B line that contains an X-ray nanoprobe on microXRF (micro X-ray fluorescence) mode. We also performed single cell analysis of the elemental distribution upon acute ionizing radiation exposure ESRF, France
