I consider myself a hardworking person, whose main areas of interest are Microbiology, Molecular Biology, Immunology and Antibiotic Resistance, with a view to applied drug development and Public Health. I obtained a bachelor's degree in Cellular and Molecular Biology by FCT-UNL in 2018, with a final classification of 16/20, after I successfully presented my bachelor project "Construção de um mutante knockout do namH em Mycobacterium smegmatis e construção de um plasmídeo para expressar a proteína NamH". After completing my bachelor internship at the Mycobacterial Pathogenesis lab (Prof. Maria João Catalão, Faculty of Pharmacy), my interest in mycobacteria as unique and extremely complex living beings led me to pursue my master studies in this area. Tuberculosis, the deadliest infectious disease in the world, which is estimated to kill 1.5 million people annually, is a global health emergency due to the emergence of multidrug, extensively-drug and totally-drug resistant strains of Mycobacterium tuberculosis. The peptidoglycan (PG) layer of the mycobacteria is specifically relevant since it features unique modifications, of which the N-glycolylation of PG muramic acid, catalyzed by the activity of the N-acetyl muramic acid hydroxylase (NamH), is of special interest. Therefore, my master thesis was focused on studying the role of the N-glycolylation of PG in host immune recognition and in antibiotic resistance. To do so, I have constructed several namH knockdown mutants in both M. smegmatis and Mycobacterium tuberculosis H37Ra using CRISPR interference. Through hard-work and perseverance, I have optimized various experimental procedures and obtained some interesting results in the scope of antibiotic resistance and host-pathogen interactions during my master thesis. I presented and discussed my dissertation on the 4th of January of 2021, having obtained the final classification of 19/20. As a result, I have completed the master course of Biopharmaceutical Sciences by the Faculty of Pharmacy of the University of Lisbon in January 2021, with a final classification of 19/20 and one Merit Award. In the meantime, I have remained at the same laboratory as a Research Collaborator and started to investigate the potential activity of NamH as a Metallo-ß-lactamase, as hypothesized in my master's dissertation. Currently, I am pursuing a Ph.D. in Pharmacy, with a specialization in Microbiology. During the Ph.D. program, I plan to continue my previous lines of research. I intend to further the study of the relevance of the unique modifications of mycobacterial peptidoglycan (N-glycolylation of muramic acids, amidation of D-isoglutamate and amidation of m-DAP) in the antibiotic resistance and in the host-pathogen interactions of Mycobacterium tuberculosis. In the future, I will work to achieve my goals, which include having professional experience abroad and teaching.
