Benedita Pinheiro

I am a researcher in at the Applied Molecular Biosciences Unit & i4HB Associated Laboratory-NOVA School of Science and Technology (UCIBIO-FCT-NOVA). I’m a member of the UCIBIO Macromolecular Crystallography and Functional Glycobiology Labs. I have been responsible for applying structural and sequence bioinformatics for the identification, construct design and recombinant expression of carbohydrate-binding and carbohydrate-active proteins. I have specialized in the analysis of these proteins using glycan-microarrays, x-ray crystallography and several biophysical characterization techniques such as Isothermal titration calorimetry (ITC) and Microscale Thermophoresis (MST). I also give assistance in molecular biology in both groups. PHD TRAINING My PhD was focused on the study at the molecular and atomic level of the cohesin-dockerin interaction, important for the assembly of a multi-enzymatic complex involved in the degradation of the plant cell wall by cellulolytic bacteria, the cellulosome. For this, I gained expertise in Molecular Biology essential for the recombinant production of the cohesin and dockerin proteins and mutational analysis of the function. In the initial study, I specially devised and implemented an essay using Non-Denaturing Gel Electrophoresis (NGE) for the study of cohesin-dockerin pairs and complemented with a thermodynamic analysis of the interaction by Isothermal Titration Calorimetry (ITC) and structurally characterization by X-ray Crystallography. In my PhD, I was also involved in the cloning and production of other protein modules constituents of the C. thermocellum cellulosome like esterases, glycohydrolases and carbohydrate-binding modules (CBM). POST-DOC TRAINING For my Post-Doc, I decided to deepen my knowledge in X-ray Crystallography moving to the group of Prof Maria João Romão. During this time, I initiated a collaboration with Dr Angelina Palma, having been introduced to glycan microarrays. I have also received training in HTP techniques for protein production (EMBO course) and Structural Bioinformatics (EMBL-EBI course). RESEARCH CONTRIBUTIONS I was involved in setting up molecular biology techniques and protocols in the Macromolecular Crystallography laboratory, which opened new avenues of research. I implemented the protein microarray technology in the Functional Glycobiology laboratory within the scope of a project grant funded by FCT to establish the proof-of-concept. I have been involved in several funded projects (2022.06104.PTDC, PTDC/BIA-MIB/31730/2017, PTDC/BIA-MIB/31730/2017, PTDC/QUI-QUI/112537/2009) where X-ray crystallography is combined with glycan microarray analysis, revealed the unique selective binding of several carbohydrate interacting proteins from bacteria from the rumen and from the human gut. I’m currently part of the internationally funded project GLYCOTwinning (HORIZON-WIDERA-2021-101079417). SUPERVISION, I have been the supervisor of three MSc student Thesis and was involved in the training of four PhD students, 3 MSc researchers and 7 undergraduate students. I have been involved in the tutoring of students for the program Verão com Ciência. TEACHING I have given regular support to Biochemistry and Structural Biology classes. I annually teach practical classes for the subject “Metabolismo e Regulação”. OUTREACH I participate in the FCT Open day and European Researcher Night.
