Over the years, Dr. Carvalho has studied the role of adipocytes in the early development of insulin resistance, obesity, diabetes and its complications. She has studied insulin resistance (primarily in young first-degree relatives (FDRs) of people with type 2 diabetes. The crosstalk between adipocytes and other organs is particularly important in populations at risk for diabetes, starting early in life. Some of these studies were performed during her PhD, in Prof. Ulf Smith¿s laboratory, at the Lundberg Laboratory for Diabetes Research, Gothenburg, Sweden. She has discovered that low insulin receptor substrate-1 protein levels in adipose tissue are predictors of pre-diabetes/diabetes. During her Post-Doctoral work, under the supervision of Dr. Barbara Kahn, at Harvard Medical School in Boston, she focused on characterizing mice lacking glucose transporter 4 (Glut4) selectively in muscle and overexpressing Glut4 only in adipose tissue to determine whether overexpression of Glut4 in adipose tissue could rescue/reverse the insulin resistance in muscle-Glut4 knockout mice. She observed that over-expressing Glut4 in adipose tissue completely reversed the insulin resistance observed in muscle-specific Glut4-deficient mice. More recently, she is assessing how pharmacological prescription drugs, particularly antipsychotic and immunosuppressive drugs, can contribute to insulin resistance development. Moreover, she has also been interested in studying the complexities of one of the most severe complications of diabetes, wound healing and tissue regeneration of the skin, with the goal of enhancing early diagnosis of subjects at risk for this chronic complication, in order to improve/personalize treatment. In this context she has aimed at evaluating the role of microRNAs and antimicrobial peptides in animal models of diabetic wound healing and infection as well as in humans with chronic diabetic foot ulcers. Over the years, she has supervised many students, including Master¿s, PhD¿s and post-doctoral fellows, from several countries, including Erasmus students. She has published over 80 peer-reviewed publications and has managed to secure several million Euros in funds from both national and international foundations, including the European Diabetes Foundation, the Diabetes UK and the National Institutes of Health. She is ad hoc reviewer for over 35 Scientific Journals. In addition, Dr. Carvalho is a Grant reviewer for Diabetes UK, MRC, Wellcome Trust, ANR-France, FNP-Poland, FWF-Der Wissenschaftsfonds, Fonds de Recherche du Quebec ¿ CONACYT, Fund for Scientific and Technological Development (FONDECYT) - CHILE. She is a Senior Editor for the Journal of Endocrinology and the Journal of Molecular Endocrinology, and more recently she was invited to become a board member of the European Tissue Regeneration Society. For many years she has been participating in science and health outreach, that aims at promoting public awareness and understanding of science and health issues especially diabetes education, teaching both adults and children in the public setting at schools, health fairs, churches and universities, in Portugal, Guinea-Bissau and in the USA.

  1. Cyclosporine A and tacrolimus reduce the amount of GLUT4 at the cell-surface in human adipocytes: increased endocytosis as a potential mechanism for the diabetogenic effects of immunosuppressive agents. Context:Immunosuppressive agentes are associated with profound metabolic side effects including new-onset diabetes and dyslipidemia after organ transplantation. Objective: Toi nvestigated the effects of the cyclosporine A(CsA)or tacrolimus ong lucose uptake and insulin signalling in human adipocytes and their impact on the regulation of celular trafficking of the glucose transporter 4 (GLUT4). 2. Neurotensin-loaded collagen dressings reduce inflammation and improve wound healing in diabetic mice. Impaired wound healing is an important clinical problem in diabetes mellitus and results in failure to completely heal diabetic foot ulcers (DFUs), which may lead to lower extremity amputations. In the present study, collagen based dressings were prepared to be applied as support for the delivery of neurotensin (NT), a neuropeptide that acts as an inflammatory modulator in wound healing. 3. Role of Endothelial Progenitor Cells and Inflammatory Cytokines in Healing of Diabetic Foot Ulcers. Background: To evaluate changes in endothelial progenitor cells (EPCs) and cytokines in patients with diabetic foot ulceration (DFU) in association with wound healing. Methods: We studied healthy subjects, diabetic patients not at risk of DFU, at risk of DFU and with active DFU. We prospectively followed the DFU patients over a 12-week period. We also investigated similar changes in diabetic rabbi. 2013. 4. mTOR inhibition with rapamycin cause impaired insulin signalling and glucose uptake in human subcutaneous and omental adipocytes. Rapamycin is an immunosuppressive agent used after organ transplantation, but its molecular effects on glucose metabolism needs further evaluation. We explored rapamycin effects on glucose uptake and insulin signalling proteins in adipocytes obtained via subcutaneous (n = 62) and omental (n = 10) fat biopsies in human donors. At therapeutic concentration (0.01 lM) rapamycin reduced basal and insul. 2012. 5. Improved Survival, Vascular Differentiation and Wound Healing Potential of Stem Cells Co-Cultured with Endothelial Cells. In this study, we developed a methodology to improve the survival, vascular differentiation and regenerative potential of umbilical cord blood (UCB)-derived hematopoietic stem cells (CD34+ cells), by co-culturing the stem cells in a 3D fibrin gel with CD34+-derived endothelial cells (ECs). ECs differentiated from CD34+ cells appear to have superior angiogenic properties to fully differentiated ECs. 2011. Pedroso, Dora C.
