Brandon Aguiar completed his Bachelor degree in Chemistry in 2016 at the University of Porto, Faculty of Sciences under the guidance of Prof. Fernanda Borges and his M.SC degree in Analytical, Clinical and Forensic Toxicology at the University of Porto, Faculty of Pharmacy under the guidance of Prof. Isabel Almeida and Prof. Helena Carmo. While working in his Bachelor's degree project " Development of new antioxidants compounds using natural models" Brandon was responsible for synthesize, purify and characterize by Nuclear Magnetic Resonance spectra (1H, 13C NMR and DEPT) new antioxidants based on ferulic acid scaffold and later evaluate their antioxidant activity using DPPH and ABTS radicals assay. During his Master's thesis Brandon was responsible for obtaining and analyzing UV/VIS spectra and implementing a ROS generation protocol to evaluate photoreactivity of chemicals by spectrophotometric monitoring the generation of singlet oxygen and superoxide anion. He also acquired experience in statistical analysis using One-way ANOVA to interpret the obtained results and became especially proficient in in vitro cell methodologies including cytotoxicity and phototoxicity evaluation using a neutral red assay in the HaCat cell line. He was also responsible for organizing and planning experimental work and for lab maintenance. In 2019, Brandon worked in a research project focused on the development of centrally-active and safe catechol O-methyltransferase inhibitors where he was responsible for synthesize, purify and characterize by Nuclear Magnetic Resonance spectra (1H, 13C NMR and DEPT) new antioxidants based on kojic acid and deferiprone scaffolds. While working on this project he developed very crucial skills in organic synthesis such as Microwave-assisted Cu-Catalyzed Protodecarboxylation of aromatic carboxylic acids, Jones Oxidation, protection of reactive groups, amidations and various purification techniques such as liquid-liquid extraction, filtration under reduced pressure and purifications through column chromatography on silica gel by classical method and by Biotage IsoleraTM Prime. In 2020 Brandon worked as a research fellow under the guidance of Prof. Fernanda Borges in the project: Redox-active phytochemicals triggering mitochondrial hormesis: developing a new generation of cosmetic active ingredients where he further improved his skills in organic synthesis, purification and characterization. Currently, Brandon is working on his PhD in Pharmaceutical Sciences with a scholarship funded by FCT in the project intilted: ROS-sensitive nanoparticles as carriers for Edaravone delivery: an innovative strategy in Amyotrophy Lateral Sclerosis treatment. In his PhD, Brandon acquired experience in the synthesis and characterization (DLS, HPLC, Release Studies, ABTS assay) of polymeric, hybrid and lipidic nanoparticles as well as in cell culture (SH-SY5Y, HepG2, NSC-34 and Caco-2 cell line) and in In vitro studies including Neutral Red, Razasorine, ROS and sulforhodamine B (SRB) assay.
