Bruno Manadas has a degree in biochemistry and a PhD in Cell Biology. He became full PI at CNC-UC in 2017, leading a team with background in medicine, psychology, biochemistry, chemistry, biotechnology, and pharmaceutical biotechnology. He has over 120 publications, 3 projects from FCT as PI and 5 as co-PI, is the head of the Mass Spectrometry node at CNC of the National Mass Spectrometry Network since 2006, is an invited peer-reviewer of over 20 journals, Associated Editor of Frontiers in Molecular Neuroscience, and Frontiers in Psychiatry, Member of the European Proteomics Association (EuPA) education committee since 2021, and board member of the Portuguese Proteomics Association (Procura). Bruno Manadas was also enrolled in the settlement of the central hospital biobank with his expertise on sample storage methods for large screening approaches. His academic activities also include being responsible for the curricular units of Neurobiology, Biology of Aging, Proteomics, and Molecular Diagnosis in the last 10 years at the Portuguese Catholic University; and responsible for advanced courses at the CNC PhD program (one sponsored by the European ESFRI Instruct network). His major focus has been on elucidating cell biology mechanisms and translational biomarker research using mass spectrometry-based approaches. One example of a cell biology research was the identification of cofilin, through proteomics screenings, and its validation as a central protein in the mechanomodulation mechanisms of mesenchymal stem cells, co-headed with Dr Mario Graos (Domingues, C., ...., B. Manadas* and M. Graos* (2020). "Cofilin-1 Is a Mechanosensitive Regulator of Transcription." Front Cell Dev Biol 8(678): 678. DOI 10.3389/fcell.2020.00678). The identification of new biomarkers for Parkinsons disease using in vitro cellular secretome proteomics screening and validation of the biomarkers in plasma samples from PD patients is one example of translational biomarkers research being performed in his lab (Anjo, S. I., ...., B. Manadas (2020). "A different vision of translational research in biomarker discovery: a pilot study on circulatory mitochondrial proteins as Parkinson's disease potential biomarkers." Transl Neurodegener 9: doi: 11.10.1186/s40035-020-00188-0). Bruno Manadas has also been coordinating the development of new MS-based approaches, p.e., to study protein oxidation and its relevance in cell biology mechanisms and biomarkers in oxidative stress conditions (Anjo, S. I., ..., B. Manadas (2019). "oxSWATH: An integrative method for a comprehensive redox-centered analysis combined with a generic differential proteomics screening." Redox Biol 22: 101130. DOI 10.1016/j.redox.2019.101130). Bruno Manadas also has experience in student supervision with 2 concluded Ph.Ds, 3 ongoing PhD students as supervisor, 4 ongoing PhD students as co-supervisor, and 23 concluded Master thesis. Current funding includes a CoLab(AccellBio), with a research contract and 100k€ for consumables, a Santa Casa da Misericórdia project as co-PI, and services to support ongoing and future PhD projects.

  4. Co-Transplantation of Adipose Tissue-Derived Stromal Cells and Olfactory Ensheathing Cells for Spinal Cord Injury Repair. Patients suffering from spinal cord injury (SCI) still have a dismal prognosis. Despite all the efforts developed in this area, currently there are no effective treatments. Therefore, cell therapies have been proposed as a viable alternative to the current treatments used. Adipose tissue-derived stromal cells (ASCs) and olfactory ensheathing cells (OECs) have been used with promising results in di. 2018. 5. Influence of passage number on the impact of the secretome of adipose tissue stem cells on neural survival, neurodifferentiation and axonal growth. Mesenchymal stem cells (MSCs), and within them adipose tissue derived stem cells (ASCs), have been shown to have therapeutic effects on central nervous system (CNS) cell populations. Such effects have been mostly attributed to soluble factors, as well as vesicles, present in their secretome. Yet, little is known about the impact that MSC passaging might have in the secretion therapeutic profile. O. 2018. 6. Secretome of Undifferentiated Neural Progenitor Cells Induces Histological and Motor Improvements in a Rat Model of Parkinson's Disease. Parkinson's disease (PD) is a progressive neurodegenerative movement disorder that results from the death of dopamine (DA) neurons. Over recent years, differentiated or undifferentiated neural stem cells (NSCs) transplantation has been widely used as a means of cell replacement therapy. However, compelling evidence has brought attention to the array of bioactive molecules produced by stem cells, d. 2018. 8. Mesenchymal Stem Cell Secretome: A Potential Tool for the Prevention of Muscle Degenerative Changes Associated With Chronic Rotator Cuff Tears. Background: Massive rotator cuff tears (MRCTs) are usually chronic lesions with pronounced degenerative changes, where advanced fatty degeneration and atrophy can make the tear irreparable. Human mesenchymal stem cells (hMSCs) secrete a range of growth factors and vesicular systems, known as secretome, that mediates regenerative processes in tissues undergoing degeneration. Purpose: To study the e. 2017. 9. Impact of the Secretome of Human Mesenchymal Stem Cells on Brain Structure and Animal behavior in a Rat Model of Parkinson’s Disease. Research in the last decade strongly suggests that mesenchymal stem cell (MSC)-mediated therapeutic benefits are mainly due to their secretome, which has been proposed as a possible therapeutic tool for the treatment of Parkinson's disease (PD). Indeed, it has been shown that the MSC secretome increases neurogenesis and cell survival, and has numerous neuroprotective actions under different condit. 2017. 10. Crosstalk between glial and glioblastoma cells triggers the "go-or-grow" phenotype of tumor cells. Background: Glioblastoma (GBM), the most malignant primary brain tumor, leads to poor and unpredictable clinical outcomes. Recent studies showed the tumor microenvironment has a critical role in regulating tumor growth by establishing a complex network of interactions with tumor cells. In this context, we investigated how GBM cells modulate resident glial cells, particularly their paracrine activi. 2017. 11. Impact of mesenchymal stem cells' secretome on glioblastoma pathophysiology. Background: Glioblastoma (GBM) is a highly aggressive primary brain cancer, for which curative therapies are not available. An emerging therapeutic approach suggested to have potential to target malignant gliomas has been based on the use of multipotent mesenchymal stem cells (MSCs), either unmodified or engineered to deliver anticancer therapeutic agents, as these cells present an intrinsic capac. 2017.
